In 2021, the European Food Safety Authority concluded that titanium dioxide is no longer safe in foods due to the same concerns over nanoparticles. As a result, titanium dioxide is now banned as a food additive in the EU. Although studies have shown that the absorption of ingested titanium dioxide is low, evidence suggests that titanium dioxide nanoparticles can accumulate in the body over time. Health Canada deemed it safe in 2022 but noted concerns. Unlike their European counterparts, Canadian officials did not consider studies performed with titanium dioxide nanoparticles alone. 

Less frequently, we ingest E171 through liquids such as salad dressing, dairy products, and some artificially colored drinks. However, since E171 is insoluble, manufacturers must use other stabilizers to keep E171 suspended in liquids as an emulsion; otherwise, it will settle to the bottom. 

The Scientific Committee on Consumer Safety issued an opinion of the safety of titanium dioxide in food, stating that it should no longer be considered as safe when used as a food additive.

While IARC listed titanium dioxide as “possibly carcinogenic to humans,” they also add that “there is inadequate evidence in humans for the carcinogenicity of titanium dioxide.” Of the four human studies that they reviewed, only one showed a potential risk for occupational workers inhaling titanium dioxide particles and lung cancer, while the other three showed no risk for cancer at all. And it’s key to note that IARC did not assess the effects of titanium dioxide found in foods.

Variations of titanium dioxide are added to enhance the whiteness of paint, plastics, and paper products, though these variations differ from the food-grade ones for things we eat (1Trusted Source, 2Trusted Source).

The compound in the Asia Pacific market experienced a mixed trends throughout the fourth quarter of 2021. These market attitudes were mostly related to lower operational loads at Chinese domestic manufacturing plants.

Over the last several years, nanoparticles have come under scrutiny for adverse health effects. Nanoparticles are ultrafine particles between 1 to 100 nanometers in diameter. (To put this in perspective, the average human hair is around 80,000 nanometers thick.) Because of their size, which can be engineered and manipulated at the atomic or molecular level, nanoparticles exhibit unique physical, chemical, and biological properties. Titanium dioxide is one of the most commonly produced nanoparticles in the world.

Thermogravimetric analysis (TGA) was conducted in a sample of vitaminB2@P25TiO₂NPs using a TA-THA Q5000 equipment. Temperature ramp rate: 10 °C/min, maximum temperature: 1000 °C, under air. Part of the same sample was mounted on conductive copper tape grids and observed through a Carl Zeiss Sigma scanning electron microscope (SEM) with an EDS probe, at the “Laboratorio de Microscopía y Análisis por Rayos X” (LAMARX) of National University of Córdoba (Argentina).

In a study published in the journal Toxicology, researchers examined the effects of exposing human colon cancer cell line (HTC116) titanium dioxide food additives in vitro. “In the absence of cytotoxicity, E171 was accumulated in the cells after 24 hours of exposure, increasing granularity and reactive oxygen species, inducing alterations in the molecular pattern of nucleic acids and lipids, and causing nuclei enlargement, DNA damage and tubulin depolymerization,” the scientists wrote. Researchers removed the additive from the culture, then examined the results 48 hours later. They found, “The removal of E171 was unable to revert the alterations found after 24 h of exposure in colon cells. In conclusion, exposure to E171 causes alterations that cannot be reverted after 48 h if E171 is removed from colon cells.”

As of August 7, the use of titanium dioxide in food is banned in the European Union. Europe is taking a precautionary principle approach based on findings from the European Food Safety Authority (EFSA).

What is an exposure route? 

When we purchase lithopone, we must pay attention to its ratio. This can be seen to some extent from the appearance. Basically, we can see that good  products are very delicate. , and the color is also very uniform, a kind of shiny white, while inferior lithopone has uneven particles and wrong luster.

Additionally, the committee noted that the available data did not provide convincing evidence of genotoxicity for titanium dioxide as a food additive, but recognized the limitations in current methodologies with respect to the testing of poorly soluble particulate materials. Although there were uncertainties in the genotoxicity data, the experts took into account the fact that the additive was not carcinogenic in adequately conducted two-year studies in mice and rats at doses of up to 7,500 mg/kg BW per day for mice, and 2,500 mg/kg BW per day for rats, the highest doses tested. There was also no evidence of reproductive or developmental toxicity in studies in rats at doses up to 1,000 mg/kg BW per day, the highest doses tested.  

French researchers studied how and where E171 nanoparticles enter the bloodstream, first studying the route through pigs and then in vitro with human buccal cells, for a 2023 study published in the journal Nanotoxicology. The research showed that the nanoparticles absorbed quickly through the mouth and then into the bloodstream, before damaging DNA and hindering cell regeneration.

In a 2017 study published in Scientific Reports, researchers exposed rats to human-relevant levels of E171 to examine the effects of intestinal inflammation and carcinogenesis. They saw that “a 100-day E171 treatment promoted colon microinflammation and initiated preneoplastic lesions while also fostering the growth of aberrant crypt foci in a chemically induced carcinogenesis model.” They continued: “Stimulation of immune cells isolated from Peyer’s Patches [which are clusters of lymphoid follicles found in the intestine] showed a decrease in Thelper (Th)-1 IFN-γ secretion, while splenic Th1/Th17 inflammatory responses sharply increased,” researchers wrote. “A 100-day titanium dioxide treatment promoted colon microinflammation and initiated preneoplastic lesions.” The scientists concluded: “These data should be considered for risk assessments of the susceptibility to Th17-driven autoimmune diseases and to colorectal cancer in humans exposed to TiO2 from dietary sources.”
